depletion of cofactors... eg if you took something that depleted vit C and ended up with signs of scurvy and poor iron status
i ended up with dangerously low magnesium levels after taking 4000 IU of vit d3 for a couple of years (and in fact two short ie 8-10 day each megadoses, each designed to boost serum levels 50 nmol/l).

eventually my symptoms were so bad i thought i would die. i organized all my stuff for the benefit of ppl that would have to deal with my space when i was found dead.
a pharmacist set me straight when i was in for a chest xray to try to figure out why i couldn't breathe (in hindsight, spastic diaphragm), and had stopped in to the pharmacy when leaving, to ask a question more related to a connection dealing with cancer.
pharmacist ignored the cancer query, asked me about three questions, then told me to ensure i took more magnesium and timed it properly in relation to the d3 intake. 2 days later, symptoms were noticeably clearing. upon testing, reversibly. easy to make all issues come back.
took YEARS to get tissue mag stores back up. and identify which supplemental forms were worth my time. (whole other adventure with a mag product which was *too* potent).

another TiMS member was still mag deficient after a whole year of taking 10K IU d3 and magnesium daily. d3 was too high, mag either the wrong form, poorly timed, and/or not enough. he suffered terribly with horrific spasticity and ultimately ended his life a few years ago.

Yes indeed, magnesium intake is very important to be managed carefully when on high-dose D3. I am on the protocol for 2 years and a half now and been taking 1000mg mag every day to counter balance the depletion. My levels are all normal though.
