• We investigated the role of p-SIRT1 and H3K9me3 as biomarkers in multiple sclerosis.
• P-SIRT1 was found to be a biomarker of disease activity.
• H3K9me3 was also found to be a biomarker of clinical activity.
• H3K9me3 was found to be a biomarker of response to glatiramer acetate treatment.
